福氏痢疾菌膜成分与细菌抗原性和毒力的关系 被引量:1

MEMBRANE COMPONENT OF SHIGELLA FLEXNERI AND THEIR RELATIONSHIP TO BACTERIA ANTIGENICITIES AND VIRULENCES

摘要 应用免疫转移技术,用从感染福氏痢疾菌的病人获取的恢复期血清分析了福氏痢疾菌膜成分与细菌抗原性和毒力的关。发现福氏痢疾菌的膜蛋白67kD 和63kD 均含有两种成分,一种和膜蛋白60kD 都可能是保护性抗原,而另一种与膜蛋白78kD 和35kD一样与福氏痢疾菌的毒力有关。采用微细胞同位素掺入示踪显示这些与病人恢复期血清反应的膜蛋白由质粒编码。 Our previous works have shown thatmembrane proteins of S.flexnert can elicitprotection against the bacteria in vivo.In thisstudy,we further,using human convalescentsera (HCS) from adult patients infected withS.flexneri,investigated the relationship be-tween membrane components of S.flexneriand their antigenicities and virulences.It wasshown in immunobloting that 5 virulent stra-ins of S.flexneri and avirulent strain T32derived from S.flexneri 2a by Istrati in Ro-mania and used as a live vaccine,had threecommon proteinsbands formed by the compo-nents of MW 67kD,63kD and 60kD witheither HCS from patients infected with S fle-xneri 2a and 3a or HCS absorbed by an avi-rulent variant strain 24570 of S.flexneri 2a,while virulent strains M90T and M25—8A(pWR110)minl shown another two bands ofMw 78kD and 35kD respectively.In con-trast,no band was found with strain 24570.HCS from patients infected with S.flexnerila,2a and 3a as well as those HCS absorbedby 24570 had blocking effect on the invasi-veness of virulent strain of S.flexneri to thekerato conjunctiva of guinea pigs;neverther-less,HCS absorbed by T32 lost the blockingeffect accompanied by disappearance of 67kD,63kD and 60kD components when T32 wasused as antigen indicating that 67kD,63kDand 60kD membrane components of S.flex-neri may accout for the antigenof S.flexnerieliciting protection antibodies in vivo.Howe-ver,HCS absorbed by T32 did still bind thecomponents 67kD,63kD and 78kD,35kD ofM90T and F2a-12,suggesting that in 67kDand 63kD bands,there exist two kinds ofcomponents with same molecular weight butdifferent antigenicities,one may accout forthe protection antigens and the other,toge-ther with 78kD and 35kD components,cor-responded to deierminants of the virulence ofS.flexneri.Further studies,using minicellcultivation and autoradiography demonstratedthat the membrane components reacting withthe HCS in the virulent strains of S.flexneriare coded by plasmid.
