摘要
应用RP-HPLC方法检测血清中氟脲嘧啶(Fluorouracil)浓度,本方法采用C18分析柱及预柱,流动相为甲醇-0.5%乙酸水溶液(5:95v/v),检测波长266nm,血清样品用乙酸乙酯萃取,氮气吹干,血药浓度在25ng~40μg/ml范围内呈线性关系,相关系数0.9999,最低检测浓度为25ng/ml。生物样品的方法回收率为101.2%~107.7%,日内日间变异系数在1.13%~6.24%(n=5)。
A method was developed for fluorouracil concertration in human serum by RP-HPLC. The analytical and preparing column was filled with a c18 stationary phase,the mobile phase consisted of a mix-ture of methanol-0. 5% acetic acid solution (5: 95v/v). The detective wavelength was 266nm. The fluo-rouracil in human serum can be extracted well with ethyl acetate and be blowed dry with N2. The calibra-tion curve was linear in the range from 0.025 to 40 μg/ml (r = O. 9999), the limit of determination was 25ng/ml;the rate of recovery 101. 2 % to 107. 7 %. the RSD within day and between day were lower than 6.24% (n = 5).
