以结膜巨噬细胞作为抗原呈递细胞诱导实验性过敏性结膜炎 被引量:1

摘要 目的:过敏性结膜炎(AC)是发生在眼部的超敏反应,然而抗原呈递细胞(APCs)参与实验性过敏性结膜炎的发展过程,APCs接受,加工处理抗原并呈递给T细胞,探讨和研究此抗原呈递细胞的类型。方法:对特异性卵清蛋白(OVA)T细胞受体转基因老鼠(DO11.10)进行处理,对两组实验鼠分别进行结膜下注射磷酸盐缓冲液(PBS)和卵清蛋白-金(OVA-gold),24h后获取结膜组织进行免疫组化分析和光镜检测其包裹OVA-gold的细胞显型。结果:结膜下注射OVA-gold的DO11.10鼠的结膜出现大量的嗜酸性粒细胞和CD4+T细胞,与结膜下注射PBS的对照组比较,差异具有统计学意义(P<0.01),接受包裹OVA-gold的抗原呈递细胞表达分化CD68,主要组织相容性复合体(MHCII)。结论:结膜巨噬细胞是实验性过敏性结膜炎发展过程中的抗原呈递细胞。 Objective:To investigate and analyze the type of antigen(Ag)-presenting cells(APCs).Methods:Ovalbumin(OVA)-specific T cell receptor transgenic(DO11.10) mice were divided into two groups.DO11.10 mice were subconjunctivally injected with PBS,OVA-gold.24h later,the conjunctival tissue was harvested for immunohistochemistry analysis and light microscopy to identify cells that engulfed OVA-gold.Results:Subconjunctival injection of OVA-gold induced infiltration of eosinophils and CD4-positive T cells into the conjunctiva,with a significant difference.The cells that took up OVA-gold into their cytoplasma expressed cluster of differentiation CD68 and major histocompatibility complex(MHC) class II.Conclusion:Conjunctival macrophages are APCs during the development of experimental AC.
