摘要
本文报告了巴氏甲烷八叠球菌 BTC 菌株的鉴定结果。该菌株在生活周期内均呈大小不等的团块聚集,8个不规则的球形细胞聚集在一起是团块构成的基本单位,未见单细胞阶段。在紫外线照射下,菌落荧光为蓝绿色,菌体荧光为蓝白色,活细胞或干制标本均可发出荧光。在以甲醇为生长基质时,其倍增时间为9.85h。可以利用甲醇、乙酸钠、三甲胺及 H_2/CO_2为碳源;不能利用甲酸、乙醇、丙醇和丁醇等。最适生长 pH 为7.0。DNA 的 G+c mol%含量为41.5%。该菌株与巴氏甲烷八叠球菌近似或相同,而与马氏甲烷八叠球菌不同。其倍增时间较巴氏甲烷八叠球菌227菌株短,DNA 中 G+c mol%含量也与已报道过的各菌株有所差别,所以定名为巴氏甲烷八叠球菌 BTC 菌株(Methanosarcina barkeri BTC)。
A strain of Methanosarcina barkeri wasfurther characterized.The strain tends toform lumps with eight irregular coccoid cells.Single cell was never found.The fluores-cence of its colony was dark-blue and of itscell was light-blue.Both viable and dry cellsare fluorescent under ultraviolet light.Me-thanol,acetate,trimethylamine and H_2/CO_2were utilized as carbon sources.Formate,ethanol,propionol and butynol could not bemetabolized.Its optimum growth pH andtemperature are 7.0 and 35℃,respectively.Its doubling time is 9.85h with methanol assubstrate,which is shorter than of M.barkeristrain 227.The G+C mol% is 41.5%,slightlydifferent from that of any other reportedstrains of Methanosarcina.Therefore,thisstrain is identified as a new strain of M.barkeri and is named strain BTC.
