摘要
目的:研究虫草肾茶胶囊对培养在高糖条件下人肾小球系膜细胞(HMC)转化生长因子-β1mRNA(TGF-β1mRNA)和纤维蛋白溶解酶原激活物抑制剂-1mRNA(PAI-1mRNA)表达的影响,探讨其在糖尿病肾病(DN)防治中的意义。方法:制备虫草肾茶胶囊及福辛普利药物血清,用实时定量PCR方法测定各组HMCTGF-β1以及PAI-1mRNA表达,并以福辛普利为对照。结果:虫草肾茶胶囊高、中剂量组及福辛普利组TGF-β1mRNA和PAI-1mRNA表达均低于高糖组,且以虫草肾茶胶囊高剂量组表达最低(P<0.05)。结论:高糖可促进HMCTGF-β1mRNA和PAI-1mRNA表达,虫草肾茶胶囊可下调高糖刺激下HMCTGF-β1mRNA及PAI-1mRNA的表达,从而预防肾小球硬化的发生、发展,进而延缓DN的进展。
Objective:To study the influence of Capsule of Chong-cao Shen-cha on the expressions of TGF-β1mRNA and PAI-1mRNA in anthropo-mesangial cell(HMC) under the condition of high glucose,and investigate the significance on diabetic nephropathy.Method: At first Capsule of Chong-cao Shen-cha and fosinopril serum were made.Then the expressions of HMC TGF-β1 mRNA and PAI-1mRNA were detected in each group with the method of real-time-quantitation-PCR,and fosinopril was as the control.Result: The expressions of TGF-β1mRNA and PAI-1mRNA were lower in high and medium dose groups of Capsule of Chong-cao Shen-cha and fosinopril group than in high glucose group.And the expressions of them in high dose group of Capsule of Chong-cao Shencha were the lowest(P0.05).Conclusion: High glucose can promote the expressions of HMC TGF-β1mRNA and PAI-1mRNA,and Capsule of Chong-cao Shen-cha can do down regulation on the expressions of them stimulated by high glucose.So Capsule of Chong-cao Shen-cha can prevent beforehand from the occurrence and advancement of glomerular sclerosis,and then defer the progression of DN.
