摘要
目的建立婴儿奶瓶及尿液中双酚A的固相萃取-高效液相色谱法。方法奶瓶经过一定温度浸泡,将溶出的双酚A经固相萃取,甲醇提取,优化HPLC-荧光法进行检测;用β-葡萄糖酸酐酶、硫酸酯酶水解婴儿尿液中的结合型双酚A,酶解后溶液采用固相萃取柱浓缩、富集和净化,优化HPLC-荧光法进行检测。结果方法的线性范围为0.05~1.0 mg/L,检出限为0.0003 mg/L,加标回收率为71.3%~101.2%。结论该方法测定婴儿奶瓶和尿液中双酚A,具有较高灵敏度、精密度。运用该方法进行实际样品的检测,得到一些非常有价值的科研数据。
Objective To establish a comprehensive analytical method based on high performance liquid chromatography fluorescence detection for determining BPA in infants' nursing bottles and urine. Methods The urine samples were de-conjugated by adding β-glucuronidase and sulfatase.After enzymatic treatment,the samples were absorbed by GDX-502.The nursing bottles were soaked in water in a certain temperature for some time.The BPA in water was absorbed by GDX-502.All samples were achieved by a Hypersil ODS column. Results The linear range of the calibration curve was 0.05~1.0 mg/L,with the detection limit of 0.0003 mg/L.The recovery of BPA was 71.3%~101.2%. Conclusions The results show that the method is simple,sensitive,accurate,and suitable for the determination of BPA in nursing bottles and urine.Some valuable experimental data are collected by using this detection method.
