摘要
通过Sephadex G-200柱层析纯化得到球形芽孢杆菌Ts-1毒蛋白,其中主要是42kDa和43kDa两种毒蛋白。用此毒蛋白免疫家兔获得抗血清。利用 ELISA 双抗体夹心法比较测定了三种灭蚊球形芽孢杆菌(Bacillus sphaericus)高毒株和两种无毒株,证明 ELISA 具有很强的特异性。ELISA 测定球形芽孢杆菌 Ts-1纯化毒蛋白,最低可检值为1.56×10-5mg/ml。球形芽孢杆菌 Ts-1发酵液的最低可检度为1:16000倍稀释。ELISA 测定12和24小时发酵培养的 Ts-1样品处理液中毒蛋白的含量,分别为0.049mg/ml 和o.22mg/ml,毒蛋白含量相差4.59倍。而相应的生物测定 LC50值分别为0.71 ppm 和0.154 ppm,毒力相差4.61倍。ELISA 与生物测定方法结果吻合。
Bacillus sphaericus Ts-1 Mosquito larvi-cidal toxins 42 k Da and 43 k Da were isola-ted by Sephadex G-200 chromatography.Threestrains of highly toxic B.sphaericus and twonon toxic strains were screened for toxicproteins using ELISA.The lowest detectabletoxin level was 1.56×10^(-5)mg/ml.Non toxicstrains did not produce antigens reacting toeither the 42 kDa or the 43 kDa antibodies.Ts-1 cultures were examined at 12 and24h by LC_(50) bioassay against Culex pipiens.The LC_(50)'s at 12h and 24h were 0.71 ppmand 0.154 ppm,respectively,i.e.,the toxinlevel at 24h was 4.6 times the level at 12h.ELISA tests established total toxin at 0.049mg/ml and 0.225mg/ml at 12h and 24h,respectively,confirming the LC_(50) study.
