摘要
目的检测LAMC2、KLK6、SNCG等3个基因在鼻咽癌细胞株5-8F-EGFR和肝转移亚群5-8F-H3B-EGFR中的表达情况,分析这些基因在鼻咽癌肝转移过程中所起的作用。方法采用基于SYBR GREENⅠ法的实时荧光定量RT-PCR(Q-RT-PCR)检测5-8F-EGFR和5-8F-H3B-EGFR中LAMC2、KLK6、SNCG等3个基因的表达情况。结论 LAMC2、KLK6、SNCG等3个基因在5-8F-H3B-EGFR中表达均上调,且与5-8F-EGFR比较差异具有统计学意义(P<0.05)。结论 Q-RT-PCR能够敏感、特异地检测出5-8F-EGFR和5-8F-H3B-EGFR中mRNA的表达差异,方法准确可靠,操作方便;LAMC2、KLK6、SNCG有可能成为判断鼻咽癌肝转移的潜在分子标志物。
Objective To detect the expression of LAMC2,KLK6,SNCG genes in nasopharyngeal carcinoma(NPC)cell line 5-8F-EGFR and liver metastatic subpopulation 5-8F-H3B-EGFR,and to analyze the roles of these genes in the metastasis of NPC.Methods The mRNA expression of above three genes in 5-8F-EGFR and 5-8F-H3B-EGFR was detected by SYBER GREEN Ⅰ real time fluorescent quantitative RT-PCR(Q-RT-PCR).Results LAMC2,KLK6 and SNCG genes all displayed higher mRNA expression in 5-8F-H3B-EGFR than that in 5-8F-EGFR,and the difference was significant(P0.05).Conclusion Q-RT-PCR can detect the difference in mRNA expression between 5-8F-EGFR and 5-8F-H3B-EGFR sensitively and specifically.It is accurate,reliable and convenient.LAMC2,KLK6,SNCG genes may be used as potential molecular markers for the diagnosis of NPC hepatic metastasis.
