摘要
目的:研究人肺癌、成人肺及胚肺组织基因表达谱差异,观察肺癌发生中多个基因的作用。方法:用鳞状细胞肺癌、胎儿胚肺、胸外伤部分肺切除肺组织,提取总RNA 后逆转录标记cDNA 探针,与代表588 个基因的AtlasTM cDNA 阵列杂交,灰度扫描杂交信号强弱及统计分析差异。结果:肺癌组织中cmyc、cfos 相关基因等多个较正常肺组织表达增加的基因,其中有些基因表达变化与胚胎肺组织相似。结论:多个基因的表达变化在肺癌发生中起作用,且与胚肺基因表达谱相似,而表现出恶性肿瘤发生中的“胚胎化”生物学特性。
Objective:To study the genes expression profile on the tissues of the adult lung cancer,the normal adult lung and the fetal lung, and observe the effect of the changes of gene expression in lung cancer. Methods:The total RNA were respectively extracted from the lung tissues which came from the fetal,the chest hurt and the lung cancer patients. Reverse transcriptase probed the total RNA and the probes respectively hybrided to the Atlas Human cDNA expression arrays. Results:The genes expression level,such as c myc and c fos genes,distinctly increased comparing with normal adult lung,but these changes occurred in fetal lung genes expression profile pattern. Conclusion:More than one gene contributed to adult lung cancer and the “fetalized”genes expression pattern also showed in adult lung cancer.
