实验性单纯疱疹性角膜炎的免疫组织化学研究 被引量:3

摘要 目的 研究单纯疱疹Ⅰ型病毒 (HSV 1)感染后疾病的严重性与宿主遗传因素的关系 ;分析HSV 1感染后小鼠角膜中炎症细胞的亚群分布 ;检测角膜三层细胞中病毒抗原的表达及其临床意义。方法 以单克隆抗体和常规组织学技术作免疫组织化学研究。结果 HSV 1感染后疾病取决于宿主的遗传因素 ,在角膜基质炎中 ,T细胞对引发疾病起主导作用 ,炎症细胞的数量与抗原量相关 ,病毒抗原沉积于角膜三层细胞 ,而BLAB/c中的频率 ( 71%~ 10 0 % )高于C5 7BL/ 6( 11%~ 5 6% )。结论 实验性单纯疱疹性角膜基质炎的严重性取决于宿主的遗传因素 :在引发角膜基质炎过程中 ,T细胞起主导作用 ;基质层为HSV 1最主要沉积场所 。 Objective To investigate the correlation between the severity of the disease and genetic factors of the host after herpes simplex virus 1(HSV 1)infection,to analyze the subpopulations of infiltrating cells in the corneas of inbred mice infected with HSV 1,and to determine the expression of HSV 1 antigen in the cornea and its clinical application. Methods Immunohistochemical study was made by conventional histology and monoclonal antibodies against HSV 1 antigens. Results The severity of the disease depended upon genetical host factors,T lymphocytes play an essential role in the course of stromal disease in mice.The number of inflammatory cells are correlative with the relative amount of HSV antigen in the cornea.HSV antigen and inflammatory cells were detected in all three layers of the cornea in both BALB/c and C 57 BL/ 6 mice but with high frequency in BALB/c(71%~100%)than C 57 BL/ 6(11%~55%)mice. Conclusion The severity of experimental HSK depends upon the genetic factors of the host;T lymphocytes play an essential role in the course of HSK.The stroma was the major deposit of HSV 1 antigen.Both inflammatory cells and HSV 1 antigen are coincident in frequency and locations.
