摘要
目的 观察类肝素及甘糖酯 (PGMS)对体外培养的大鼠晶状体上皮细胞 (LECs)增殖的影响。方法 将质量浓度为 0 .2 ,0 .4,0 .8mg/ml的类肝素和PGMS作用于培养细胞 ,72h后行结晶紫染色比色测定 ,并对 2 4h内的 3H TdR掺入率进行测定。结果 类肝素、PGMS可使LECs增殖明显降低 (P <0 .0 5 ) ,DNA合成明显受抑 (P <0 .0 1) ,相同质量浓度下类肝素的作用强于肝素。结论 类肝素。
Objective To study the effect of heparinoid and proglylene glycol mannate sulfate(PGMS) on the proliferation of newborn rat lens epithelial cells (LECs) in vitro. Methods LECs were exposed to 0.2,0.4,0.8*!mg/ml concentrations of heparin,heparinoid and PGMS respectively.The proliferation rate of LECs was measured by crystal violet staining and colorimetric analysis.The DNA synthesis of LECs was measured by 3H TdR incorporation. Results Heparinoid and PGMS inhibited the proliferative rate and DNA synthesis of rat LECs in vitro ( P <0.05).Heparinoid was more effective as compared to heparin at the same concentration ( P < 0.05 ). Conclusion Heparinoid and PGMS are capable of inhibiting LECs growth.These results suggest that heparinoid and PGMS could be used to prevent lens capsule opacification.
