人胚眼色素上皮细胞的培养

Culturing of human fetal retinal pigment epithelium

目的 准备可供植入的人色素上皮细胞。方法 将 12～ 2 4周人胚眼的视网膜色素上皮细胞 (RPE)自脉络膜毛细血管床上撕离 ,置入培养皿中作贴壁培养。结果 培养的RPE细胞可在培养皿中存活 2～ 3个月。顺极性和反极性培养不影响细胞的形态和存活时间。培养的RPE细胞具有吞噬视细胞外节的功能。

Objective To establish a retinal pigment epithelium(RPE)donor tissue bank for RPE allograft transplantation. Methods RPE patches were obtained from human fetal eyes aged from 12～24 weeks and cultured in regular culture dishes. Results Cultured RPE can survive 2～3 months in a culture dish either being placed right side up or up side down and maintain the typical morphological characters of epithelial cells.The cells can phagocytose the disks of rod cells. Conclusion Fetal RPE patches are excellent donor tissue for RPE allograft transplantation.
