HPLC法同时测定养血化瘀合剂中阿魏酸、甘草苷和甘草酸

Simultaneous determination of ferulic acid,liquiritin,and glycyrrhizic acid in Yangxue Huayu Mixture by HPLC

目的建立HPLC同时测定养血化瘀合剂(当归、川芎、甘草等)中阿魏酸、甘草苷和甘草酸的方法。方法采用依利特Hypersil C18(200 mm×4.6mm,5μm)色谱柱,以乙腈为流动相A,0.1%磷酸水溶液为流动相B,进行线性梯度洗脱(0～30 min,85%B→50%B),体积流量为1 mL/min,检测波长为245 nm,柱温为25℃。结果阿魏酸在14.84～148.36μg/mL(r=0.999 8),甘草苷在3.852～38.52μg/mL(r=0.999 6)和甘草酸在7.55～75.50μg/mL(r=0.999 7)的范围内浓度与峰面积呈良好的线性关系,阿魏酸、甘草苷和甘草酸的进样精密度分别为0.96%、1.02%和0.93%(n=5),测定重复性分别为1.28%、1.32%和1.26%(n=6),加样回收率分别为100.1%、100.1%和99.7%(n=9),溶液在12 h内稳定。结论本法操作简单、快速、重复性好,结果准确可靠,可用于养血化瘀合剂的质量控制。

AIM To establish a method for simultaneously determing ferulic acid,liquiritin,and glycyrrhizic acid in Yangxue Huayu Mixture（Angelicae Sinensis Radix,Chuanxiong Rhizoma,Glycyrrhizae Radix et Rhizoma,etc.）.METHODS The column was Elite Hypersil C18（200 mm ×4.6 mm,5 μm）.The mobile phase was gradient elution（0-30 min,85% B→50% B）,acetonitrile as mobile phase A,0.1% phosphoric acid as mobile phase B.The flow rate was 1 mL/min.The detection wavelength was set at 245 nm.The temperature of column was 25℃.RESULTS The linearity of ferulic acid was in the range of 14.84-148.36 μg/mL（r = 0.999 8）;that of liquiritin was in the range of 3.852-38.52 μg/mL（r = 0.999 6）;that of glycyrrhizic acid was in the range of 7.55-75.50 μg/mL（r = 0.999 7）.Their RSDs of precision were 0.96%,1.02% and 0.93%,respectively.The reproducibilities were 1.28%,1.32% and 1.26%,respectively.The average recoveries were 100.1%,100.1% and 99.7%（n = 9）,respectively.The sample solution was stable within 12 h.CONCLUSION The method is simple,reliable and reproducible.It can be used to control the quality of Yangxue Huayu Mixture.
