摘要
目的探讨内毒素(lipoplysaccharide,LPS)诱导的急性肺损伤早期肺组织肝细胞生长因子(hepatocyte growth factor,HGF)、角质细胞生长因子(keratinocyte gronth factor,KGF)的动态变化及β2受体激动剂的治疗效果。方法将78只Wistar大鼠随机分为生理盐水对照组(NS组n=6)、内毒素组(LPS组n=36)、β2受体激动剂组(n=36),向LPS和β2受体激动剂组大鼠腹腔内注射1 mL含LPS(5 mg.kg-1,n=36只)的生理盐水复制ALI模型。生理盐水对照组则给予等量生理盐水腹腔注射。β2受体激动剂组大鼠于造模30 min内经鼠尾静脉一次性给予β2受体激动剂0.02 mg.kg-1。三组实验大鼠分别于4 h、8 h、12h、16 h、24 h、32 h各随机处死6只。收集血清,冰浴匀浆肺组织制备匀浆,-80°C冻存。Elisa法测定肺组织HGF、KGF的量。计算肺组织的湿重(D)与肺组织干重(W)的比值,以此评估肺水肿的程度,进行肺组织形态学观察,并研究β2受体激动剂对肺组织匀浆液HGF、KGF的影响。结果①急性肺损伤早期肺组织匀浆液内HGF的表达呈双峰,分别于8 h、32 h达到高峰。KGF从16 h开始增高,32 h未见回落至正常水平;②β2受体激动剂干预后HGF、KGF分别在8 h、12 h升高,并持续在较高水平。结论①急性肺损伤早期大鼠肺组织HGF、KGF早期就有所升高;②β2受体激动剂能促进HGF、KGF的分泌,促进受损组织的修复。
Objective To explore the changes of hepatocyte and keratinocyte growth factors (HGF/KGF)in mice with lipopolysaccharida (LPS)induced acute lung injury and the intervention effect of β2 receptor agonist. Methods 78 wistar rats were randomly divided into saline group( n = 6 ), LPS group( n = 36 )and β2 receptor agonist group( n = 36 ). LPS group and β2 receptor agonist group rats were injected with a single intraperitoneal injection does of l mL LPS (5 mg kg-1) saline , saline group rats were injected with the same dose of saline. β2 receptor agonist group rats were injected with β2 receptor agonist 0.02 mg kg-1 by caudal vein in 30 min. 6 rats were randomly killed in each group in 4 h,8 h,12 h,16 h,24,32 h after injection. Collected the tissue homogenate, conserved in - 80℃ refrigerator. The content of hepatocyte and keratinoeyte growth factors were detected by ELISA. To observe lung tissue pathological with microscope and evaluate the pneumonedema by the ratios of wet lug tissue and dry lug tissue (W/D). Results (1) Compared with control group, the expression of hepatocyte growth factors in lung tissue homogenate with acute lung injuny exhibited a biphasic pattern and reached the peak at 8 h and 32 h respectively, and the keratinocyte growth factor in lung tissue homogenate increased at 16h and remained at a higher level at 32 h; (2)Compared with LPS group, the expression of HGF and KGF increased at 8h and 12h respectively, and still remained at higher level in β2receptor agonists group. Conclusions (1) The expression of hepatocyte and keratinocyte growth factor in β2receptor agonists group increased at early time compared with LPS group; (2) β2 receptor agonists might take effect by means of increase the expression of hepatocyte and kerati-nocyte growth factors in lung tissues, and sequentially, it promoted the restore of lung injure.
