重组日本血吸虫中国大陆株rTPI分子对小鼠免疫保护性作用的研究 被引量:12

摘要 目的研究重组日本血吸虫中国大陆株磷酸丙糖异构酶(SjC-TPI)分子抗血吸虫感染的保护性作用。方法用IPTG诱导表达,制备纯化的重组TPI蛋白,将重组TPI抗原免疫C57BL/6及昆明鼠,8周后用血吸虫尾蚴攻击感染,45d后剖杀,计数减虫率及减卵率。结果C57BL/6获得27.78%的减虫率;昆明鼠获得21.39%的减虫率,54.00%的减卵率。结论重组SjC-TPI对血吸虫感染具有一定的保护作用,可能为日本血吸虫病疫苗候选分子之一。 Objective To observe protection effect of recombinant SjC-TPI against the infec-tion of Schistosoma japonicum. Methods The recombinant SjC-TPI was expressed by IPTG induc-tion, and purified with Glutathione Sepharose 4B. The C57BL/6 and Kunming mice were immu-nized with rTPI. Eight weeks after the final immunization, the mice of immunized and control groupwere challenged with 45±1 S. japonicum cercariae. All mice were sacrificed at forty-five days afterchallenge. Results The worm reduction rate was 27.78% in C57BL/6; the worm reduction rateand the egg reduction rate were 21.39% and 54.00% in Kunming mice respectively. ConclusionRecombinant SjC-TPI has a protective effect against Schistosoma japonicum infection. It could beused as one of vaccine candidates of schistosomiasis japonica.
