大鼠单侧输尿管梗阻致肾间质纤维化与TGF β_1 mRNA表达的关系 被引量:3

摘要 目的:探讨转化生长因子β1(TGFβ1)在单侧输尿管梗阻(UUO)所致肾间质纤维化中的作用。方法:应用原位杂交,生化分析及病理形态学观察等技术,研究UUO术后1、3、7、14、21d不同时相肾皮质中TGFβ1mRNA表达与小管间质病变、羟脯氨酸代谢的关系。结果:UUO肾组织中羟脯氨酸含量逐渐增加及间质面积增宽,伴TGFβ1mRNA表达增高。结论:TGFβ1lmRNA表达增加是UUO致肾间质纤维化的重要原因之一。 Objective: To study the possible role of transforming growth factor 9, (TGF 5,) in the formation of unilateral ureteral obstruction-induced interstitial fibrosis of renal tubules in rats. Methods: Rat model of unilateral ureteral obstruction was established. In situ hybridization, biochemical analysis were used to determine the relationship of TGF 5, mRNA expression with the changes of hydroxyproline content and the morphological changes of the renal tubules, onthe 1st, 3rd, 7th, 14th adn 21st day after unilateral ureteral obstruction. ResuIts: After uni1ateral uretera1 obstruction, the content of hydroxypro1ine was significantly increased in the renal tissues and in the interstitial area of the renal tubules. Meanwhile, there was an increase of the expression of TGF 6, mRNA. ooncIusion: 1n the formation of interstitial fibrosis of the renal tubules induced by unilateral ureteral obstruction, the increase of TGF 9, mRNA expression may be one of the factors.
