正常人外周血嗜酸性粒细胞的分离与培养方法的改进 被引量:6

摘要 目的:探讨外周血嗜酸性粒细胞(Eos)纯化、分离与培养的方法。方法:采用Percoll密度梯度(1.082~1.100)分离外周血Eos,离心前细胞悬液预先与fMLP37℃孵育10min。纯化的Eos分别在加入或不加入细胞因子(IL-3、IL-5和GM-CSF)的条件下培养,观察细胞活力的变化。结果:传统Percoll密度梯度分离Eos的纯度和回收率分别为(15.8±53)%和(64.3±6.4)%。预先用fMLP孵育Eos使分离Eos纯度(96.5±1.8%)显著增高(P<0.01),回收率(68.4±71)%无明显变化(P>0.05)。Eos体外培养存活2~3d,加入细胞因子可使Eos存活时间延长6d以上。结论:fMLP可有效增加Percoll密度梯度分离Eos的纯度。分离的EoS可用于体外培养。
