多烯大环内酯类抗生素——链霉菌FR-008代谢产物的研究

Characterization of the hepaene macrolide antibiotic produced by Streptomyces sp. FR 008

链霉菌ＦＲ－００８ 是一株抗生素产生菌。发酵液经萃取、大孔树脂吸附、分配层析等方法处理，从中获得了一种七烯大环内酯类抗生素，即抗生素ＦＲ－００８。ＨＰＬＣ分析研究表明，它与灰色链霉菌ＩＭＲＵ ３５７０产生的杀假丝菌素是同一种物质，均含有４个保留时间完全相同的主要组分。其中抗生素ＦＲ－００８ｂ 经核磁共振（ＮＭＲ）研究被证实为杀假丝菌素Ｄ；ＦＲ－００８ａ也经１Ｈ－ＮＭＲ的比较分析提出了它的可能结构。

Streptomyces sp. FR 008 is a strain producing a complex of heptaene macrolide antifungal antibiotics, named FR 008, which was previously regarded to be similar to candicidin, but different in sugar moiety. The antibiotic FR 008 was extracted and separated from the cultured broth of Streptomyces sp. FR 008. A comparison between antibiotic FR 008 and candicidin has been made by studying the HPLC separation profiles of antibiotic FR 008, candicidin and a mixture of both. The retention time (RT) of each of the three main peaks of the antibiotic FR 008 is entirely identical to candicidin. The main component FR 008 was verified as candicidin D on the basis of 1 H 1H NMR and 13 C NMR studies. The possible structure of FR 008a could be proposed also after comparing the partial 1 H NMR signals of the two compounds.
