电针对脊髓损伤后硫酸软骨素蛋白多糖表达的影响 被引量:15

Effects of electro-acupuncture on expression of CSPGs after spinal cord injury

摘要 目的观察电针对脊髓损伤后硫酸软骨素蛋白多糖(CSPGs)表达的影响,探讨电针对脊髓损伤修复的可能作用机制。方法将成年SD大鼠40只分为正常对照组、模型组和电针组。建立成年大鼠T9~T10段脊髓全横断损伤模型。除电针组于术后当天进行电针治疗外,3个组均分别于术后7、14、28 d取材,用免疫组织化学ABC法和图像分析技术检测脊髓内CSPGs的表达。结果术后7~28 d,与正常对照组相比模型组脊髓损伤邻近组织内CSPGs表达明显增强(P<0.05);与模型组相比,电针组脊髓损伤邻近组织内CSPGs表达明显受到抑制,差异显著(P<0.05)。结论电针能显著降低脊髓损伤后CSPGs的表达,从而减弱CSPGs对轴突再生的抑制作用,这可能是其促进脊髓损伤修复的可能机制之一。 Objective To explore the effect of electro-acupuncture on expression of chondroitin sulfate proteoglycans(CSPGs) in the injured tissue of the transected completely spinal cord,and its mechanism in the repair of spinal cord injury(SCI).Methods Forty adult rats were divided into the normal control group,the model group and the electroacupuncture group(EC group).All rats underwent completely transection between segments T9 and T10 of the spinal cord.Electro-acupuncture treatment started on the same day after operation in the EC group.The rats were sacrificed on 7 d,14 d and 28 d after operation.Expression of CSPGs in the tissue of the injured spinal cord was determined by immunohistochemical staining.The immunopositive gray scale of the immune reactant was detected with computer image analysis technique.Results Expression of CSPGs in the injured spinal cord was apparently increased in the model group from 7 d to 28 d after operation,compared with the normal control group(P0.05).Expression of CSPGs in the injured spinal cord was remarkably inhibited by electro-acupuncture treatment,compared with the model group(P0.05).Conclusion Electro-acupuncture can significantly reduce expression of CSPGs,which is an inhibitor of axon regeneration after SCI.And this effect may be one of the mechanisms by which electro-acupuncture can promote functional recovery after SCI.
