喹啉对癫大鼠海马神经元连接蛋白36表达的影响 被引量:2

摘要 目的探讨喹啉对癫大鼠海马神经元连接蛋白36(Cx36)表达的影响。方法 64只SD大鼠随机分为正常对照组、癫模型组、地西泮治疗组和喹啉治疗组,每组16只大鼠。采用氯化锂-匹罗卡品诱导制作癫大鼠模型,地西泮治疗组予以1 mg/kg地西泮治疗,喹啉治疗组予以60 mg/kg喹啉治疗。术后分别采用Racine评分和脑电图检查判断癫发作情况。分别用免疫荧光染色法、Western blot法检测各组大鼠术后2 h、4 h时海马神经元Cx36的表达。结果与正常对照组比较,癫模型组和地西泮治疗组大鼠术后2 h、4 h时海马神经元Cx36表达水平显著升高(均P<0.01)。癫模型组和地西泮治疗组大鼠术后2 h、4 h时海马神经元Cx36表达水平比较差异无统计学意义。与癫模型组及地西泮治疗组比较,喹啉治疗组大鼠术后2 h、4 h时海马神经元Cx36表达水平显著降低(均P<0.01)。结论癫大鼠海马神经元Cx36表达水平升高,喹啉能抑制这一变化。 Objective To investigate the effect of Qunine on expression of connexin 36(Cx36) in hippocampal neurons in epileptic rats.Methods Sixty-four SD rats were randomly divided into normal control group,epileptic model group,Diazepam therapy group and Qunine therapy group,and 16 rats in each group.Epileptic rat models were made by lithium chloride-pilocarpine method.Diazepam therapy group were given 1 mg/kg Diazepam by introperitoneal injection,and Qunine therapy group were given 60 mg/kg Qunine by introperitoneal injection.The Racine score and electroencephalography were used to decide epileptic seizure after operation respectively.The expression of Cx36 in hippocampal neurons in each group at 2 h and 4 h after operation was detected by immunofluorescent staining method and Western blot method respectively.Results Compared with normal control group,the expression of Cx36 in hippocampal neurons at 2 h and 4 h after operation were obviously increased in epileptic model group and Diazepam therapy group(all P0.01).There was no significant difference of the expression of Cx36 in hippocampal neurons at 2 h and 4 h after operation between epileptic model group and Diazepam therapy group.Compared with epileptic model group and Diazepam therapy group,the expression of Cx36 in hippocampal neurons at 2 h and 4 h after operation were obviously decreased in Qunine therapy group(all P0.01).Conclusions The expression of Cx36 in hippocampal neurons may increased in epilepsy rats.In addition,Qunine can suppress this change.
