Differential Expression of Activating Transcription Factor 5 in Human Rectal Cancer

摘要 目的分析人直肠癌组织中转录激活因子5(ATF5)的表达变化及其与直肠癌临床病理特征的关系。方法选取四川大学华西医院胃肠外科中心2009年3~10月期间手术切除的直肠癌及距离肿瘤5cm以远的正常直肠组织标本92对,分别应用实时荧光定量RT-PCR和免疫组织化学染色SP法检测ATF5 mRNA和蛋白的表达情况。结果直肠癌组织中33例(35.9%)ATF5 mRNA表达上调,但ATF5 mRNA相对表达量与正常直肠组织相比,二者间差异无统计学意义(P=0.363),并且ATF5 mRNA表达与患者年龄、性别、肿瘤类型、分化程度、浸润深度、淋巴结转移、远处转移及TNM分期均无关(P〉0.05);但直肠癌组织中ATF5蛋白的表达高于正常直肠组织(P=0.000),并且其表达与肿瘤分化程度有关(P=0.013),而与其他临床病理因素无关(P〉0.05)。结论ATF5蛋白的表达与直肠癌的发生和分化相关,但这种相关需要进一步的研究证明。 Objective To determine the expression change of activating transcription factor 5 (ATF5) in hu- man rectal cancer tissue, and analyze the correlation between ATF5 expression and the clinicopathologic parameters of rectal cancer. Methods Ninety-two paired samples of rectal cancer tissue and more than 5 cm distant normal rec- tal tissue were obtained from inpatients between March 2009 and October 2009 in this hospital. ATF5 mRNA and protein expressions were detected by quantitative real-time RT-PCR and immunohistoehemieal staining. Results Thirty-three (35.9 % ) cases of rectal cancer showed ATF5 mRNA overexpression~ however, the expression level of ATF5 mRNA in the rectal cancer tissue was not statistically different from that in the normal rectal tissue (P=0. 363). There was no evidence for the relationship between the ATF5 mRNA expression and the patients' age, gender, histological type, tumor differentiation degree, invasive depth, lymph node metastasis, distance me- tastasis, or TNM stage. In contrast, the positive expression rate of ATF5 protein in the rectal cancer tissue was sig- nificantly higher than that in the normal rectal tissue (P:0. 000). Moreover, the ATF5 protein expression was cor- related with the tumor differentiation degree (P:0. 013), but not with other clinicopathologic features (P〉0.05). Conclusions The results suggest that ATF5 protein may he related to the carcinogenesis and differentiation of hu- man rectal cancer. However, further researches are required to prove the correlation.
