导流杂交基因芯片检测人乳头状瘤病毒21种亚型的应用评价 被引量:4

摘要 目的了解基因芯片检测人乳头状瘤病毒(HPV)21种亚型感染的临床应用价值。评价导流杂交基因芯片技术(凯普导流杂交)HPV DNA检测法(HybriMax)在女性生殖道HPV感染检测中的临床效用,初步探讨最常见的HPV基因型。方法利用标记有生物素的HPV通用引物进行PCR扩增,扩增产物变性后与固定在尼龙膜上21种HPV特异性分型探针进行反向斑点杂交检测HPV亚型。结果 591例患者标本中,基因芯片阳性率69.2%,其中单一亚型感染266例,重叠感染143例。共检出亚型20种。结论基因芯片一次试验可联合检测多种HPV亚型感染并分型,可研究HPV感染型别的分布,提高由HPV引起的肿瘤防治水平。 Objective To understand the clinical value of the gene chip for detection of gynecological outpatients infected with HPV 21 subtypes,and to evaluate the clinical efficacy of flow-through hybridization gene chip technology,cape flow-through hybridization HPV DNA detection method(HybriMax) in the detection of female reproductive human papilloma virus(HPV) infection,and to study the most common HPV genotypes of women reproductive tract infections.Methods Biotin labeled universal primers were used for HPV PCR amplification,PCR product was denaturized and then reversely dot-blot hybridized with 21 HPV-specific sub-type probes fixed on nylon membrane to detect HPV subtypes.Results In 591 cases of patient samples,gene-chip positive rate was 69.2%,in which 266 cases were single subtype infection,and 143 cases were coinfection.A total of 21 subtypes were detected.Conclusion The gene chip test can detect a variety of HPV subtypes infection.It is important to study the subtype distribution of HPV infection in gynecological outpatients and to improve the prevention and treatment of the cancer caused by the HPV.
