摘要
目的研究中电导钙激活的钾通道mRNA在大鼠主动脉球囊损伤后内膜增生中的作用。方法建立大鼠主动脉球囊损伤模型、高脂模型,观察假手术组、单纯球囊损伤后1,4,7,14,28 d组以及高脂、不同剂量氯沙坦下IKCa1 mRNA表达情况。结果球囊损伤后主动脉内膜增生明显,普通饮食28 d组增生达高峰;IKCa1mRNA表达升高,单纯球囊损伤后1 d组达高峰,高脂+球囊损伤时IKCa1 mRNA表达较单纯球囊损伤组1 d组增高,高剂量氯沙坦时IKCa1 mRNA表达下降。结论 IKCa1 mRNA表达增高可能在大鼠主动脉球囊损伤后内膜增生中起重要作用,IKCa1 mRNA表达的增加和下降可能是高脂血症和氯沙坦影响血管内膜增生的机制。
Objective To investigate the mRNA expression of intermediate-condutance Ca2+-activeted K+ channels(IKCa1) in rat aortic intimal proliferation after balloon injury.Methods The expression levels of IKCa1 mRNA were detected at different losartan doses and various time points(1,4,7,14,28 days after balloon injury and hyperlipidemia).Results The maximal intimal proliferation and peak intima-media ratio was found on the 28th day after balloon injury.The peak expression of IKCa1 mRNA was found on the 1st day post-injury.The expression of IKCa1 mRNA upregulation was found in the condition of hyperlipidemia and injury,and downregulation in the condition of high losartan dose.Conclusion The upregulated IKCa1 mRNA expression may play an important role in rat aortic intimal proliferation after balloon injury.Upregulation and downregulation of IKCa1 mRNA expression may be the mechanism by which hyperlipidemia and losartan act on the intimal proliferation.
