人脑胶质瘤中ING4和CTGF的表达及其意义 被引量:2

摘要 目的探讨人脑胶质瘤中生长抑制因子4(ING4)及结缔组织生长因子(CTGF)的mRNA的表达及其意义。方法采用荧光实时定量聚合酶链式反应法检测ING4和CTGFmRNA在30例脑胶质瘤组织和5例正常脑组织中的表达水平,统计分析表达水平和肿瘤级别之间的关系以及二者表达水平的相关性。结果 ING4mRNA在Ⅰ~Ⅱ级胶质瘤(低级别组)及Ⅲ-Ⅳ级胶质瘤(高级别组)中的表达均明显低于正常脑组织(P<0.01),Ⅲ~Ⅳ级胶质瘤中的表达量显著低于Ⅰ~Ⅱ级胶质瘤(P<0.01),而且随着肿瘤级别的升高,ING4的表达降低。CTGFmRNA在Ⅰ~Ⅱ级胶质瘤及Ⅲ~Ⅳ级胶质瘤中的表达均明显高于正常脑组织(P<0.01),而Ⅲ~Ⅳ级胶质瘤的表达显著高于Ⅰ~Ⅱ级胶质瘤(P<0.01),且随着肿瘤级别的升高,CTGFmRNA表达增加。通过Spearman秩相关分析,显示ING4和CTGFmRNA在胶质瘤中的表达呈负相关(γ=-0.723,P<0.01)。结论 ING4mRNA在胶质瘤组织中的表达与肿瘤的恶性程度呈负相关,CTGFmRNA在胶质瘤组织中的表达与肿瘤的恶性程度呈正相关,提示ING4和CTG可能在人脑胶质瘤的发生和发展中起重要作用。 Objective To study the expressions of ING4 and CTGF in human gliomas and their meanings.Methods The expressions of CTGF and ING4 mRNA in 30 specimens of human gliomas and 5 normal brain tissues were determined by real time polymerase chain reaction.Their relationship with pathological grades was analyzed.Results The level of ING4 mRNA expression in the human gliomas was significantly lower than that in the normal brain tissues (P0.01).The level of ING4 mRNA expression in grades Ⅲ~ Ⅳ gliomas was significantly lower than that in grades Ⅰ~Ⅱ gliomas (P0.01).The levels of CTGF mRNA expression in the human gliomas was significantly higher than that in the normal brain tissues (P0.01).The level of CTGF mRNA expression in grades Ⅲ~Ⅳ gliomas was significantly higher than that in gradesⅠ~Ⅱ gliomas (P0.01).The level of ING4 mRNA expression was negatively related to that of CTGF mRNA expression in the gliomas (γ=-0.723,P0.05).Conclusions The expression of ING4 mRNA in the gliomas is negtively related to the malignant degree of the tumors.The expression of CTGF mRNA in the gliomas is positively related to the malignant degree of the tumors.It is suggested that ING4 and CTGF may play an important role in the phatholgenesis and development of the human glioma.
