摘要
目的:探讨联合应用PTEN基因转染兔预防血管再狭窄及其发生机制。方法:建立兔腹主动脉再狭窄模型;构建PTEN表达载体并在球囊损伤腹主动脉同时转染血管壁细胞。实验分3组。对照组:单纯行内膜剥脱术;实验组A:内膜剥脱术+PTEN真核质粒;实验组B:内膜剥脱术+空脂质体。采用常规病理、免疫组织化学、以及形态测量方法观察对再狭窄血管病变的影响及血管内膜细胞增殖细胞核抗原(PCNA)和MMP-9蛋白的表达。结果:成功转染PTEN基因组显著抑制损伤后4周内膜VSMCs表达PCNA,与对照组相比PCNA表达和MMP-9的水平明显受抑制(P<0.01);显著减少血管内膜厚度和面积。结论:PTEN基因转染在一定程度上可抑制实验性兔腹主动脉再狭窄。
Objective: To discuss the effectis and mechanism of PTEN(phosphatase and tensin homolog tumor suppressor) gene on vascular restenosis in rabbits.Methods: 30 rabbits were randomly divided into three teams control group,PTEN group and empty load group fed with normal food for 4 weeks.The model of vascular restenosis of rabbit abdomen arteries was established by balloon denudation after 4 weeks.Immunohistochemistry was used to observe the protein expressions of PCNA、MMP-9.Vascular intima thickness and area was evaluated with shape measurement.Results: Compared with the control group,PTEN inhibited the protein expressions of PCNA and MMP-9 significantly(P0.01).intimal thickness(IT) and the intimal area(IA) of the PTEN group were significantly decreased(P0.01).Conclusion: PTEN gene can inhibit the proliferation of rabbit VSMCs and vascular restenosis.
