摘要
以老鹰茶为原料,槲皮素为对照品,总黄酮含量为考核指标,研究了老鹰茶总黄酮的水浴提取工艺,同时对总黄酮提取物的抗氧化活性进行了探讨。在提取温度、时间、溶剂浓度、料液比等单因素的基础上,通过正交实验,确定老鹰茶总黄酮的最佳工艺条件为:提取温度60℃,提取时间90min,乙醇浓度70%,液固比35∶1。在此条件下,测得的老鹰茶总黄酮得率为6.190%(以槲皮素计)。老鹰茶总黄酮的抗氧化活性结果表明,老鹰茶总黄酮对DPPH自由基具有较强的清除活性,其EC50值为18.17μg/mL;老鹰茶总黄酮具有较强的还原能力,其EC50值为77.52μg/mL。老鹰茶总黄酮具有较强的抗氧化活性,是一种潜在的具有抗氧化功能食品原料。
The extraction conditions of the water bath technology of total flavonoids from Litsea coreana L.was studied.The effects of temperature,extracting time,ethanol volume fraction and ratio of liquid to material on extraction effect of total flavonoids were discussed.Simultaneously,the antioxidant prosperities of total flavonoids extracts were discussed.The optimum extraction conditions of total flavonoids by using orthogonal experiment based on single factor were obtained as follows:ethanol solvents 70%,ratio of liquid to material 35 ∶1,extraction at 70℃ for 90min.Yield of rude total flavonoids is 6.190%(as quercetin).The flavonoids extracts showed more potent capacity for scavenging DPPH with EC50 value reaching 18.17 μg/L.Additionally,the flavonoids extracts exhibited higher reducing power and its EC50 value was 77.52g/L.The results indicated that the flavonoids extracts from Litsea coreana L.have strong antioxidant activities and are good ingredients of functional foods with antioxidant properties.
