摘要
根据拟南芥中参和维生素C合成和代谢关键酶的氨基酸序列,经在公共数据库中搜索比对获得了水稻中参与维生素C合成相关基因的信息。在此基础上,利用实时定量PCR技术对这些基因在水稻不同组织器官以及发育种子中的表达进行分析。结果表明:水稻维生素C合成相关基因在叶中的表达量最高,其次是叶鞘和籽粒,茎和根中的表达量最低,而幼嫩叶片中的表达水平一般高于成熟叶片。
Vitamin C, also known as L-ascorbic acid (AsA), is one of the most important and abundant antioxidant compounds that play critical roles in growth and development, as well as in protection from biotic or abiotic oxidative stresses in plants. The Smirnoff-Wheeler (SW) pathway has been proven to be the only significant source of AsA in higher plants. In this study, the coding sequences of AsA biosynthesis and metabolic related genes from Arabidopsis were used as query sequences to search in the rice genome database and we got a series of corresponding homologous genes. Real-time PCR analysis was done to examine the expression patterns of these genes in different tissues as well as in grains at different developing stages in a japonica rice. The results revealed that these genes expressed in all tested tissues, but their expression was highest in leaves, lower in developing seeds as well as in sheaths, and lowest in roots and stems. Their expressions in young leaves were much higher than in old leaves.
