摘要
利用经体外合成、用地高辛标记的阿尔巴斯白绒山羊高甘氨酸/酪氨酸角蛋白关联蛋白(HGT KAPs)基因家族9个成员的cRNA探针,通过原位杂交技术在山羊皮肤组织切片上进行定位,探测HGT KAPs基因家族9个成员在内蒙古阿尔巴斯白绒山羊10月份皮肤毛囊中的表达。结果表明:HGT KAPs在兴盛期初级毛囊和次级毛囊的皮质层均有强烈的表达信号。这说明HGT KAPs是绒山羊毛和绒皮质蛋白的组成成分,是皮质层中起角质化作用的重要蛋白质,直接参与绒毛和粗毛的合成,对毛发的理化性质具有重要作用。
cRNA probes with DIG of nine members of arbas cashmere HGT KAPs were made in vitro, and were in situ hybridized with the adult skins of arbas cashmere goat, to detect the expression of nine members of caprine HGT KAPs mRNA in the skins of Arbas cashmere goats. In situ hybridization with the goat HGT KAPs eRNA probes on skin revealed a strong positive signal in the cortex of both primary and secondary hair follicles. It demonstrated that HGT KAPs were important structure proteins of wool and cashmere cortex, and cells of cortex were keratinizated by HGT KAPs. HGT KAPs participated in the synthesis of wool and cashmere directly and affected the physico-chemical property of pelage.
