摘要
目的:检测α7烟碱型乙酰胆碱受体的不同活性状态对CD4+CD25+调节性T细胞中STAT5表达的影响。方法:免疫磁珠法分离C57BL/6J小鼠脾脏中的CD4+CD25+调节性T细胞,以烟碱/α银环蛇毒刺激后采用western blot法分析α7受体的不同活性状态对细胞中STAT5表达的影响。结果:western blot分析显示,α7烟碱型乙酰胆碱受体激动剂--烟碱可以提高调节性T细胞中磷酸化STAT5的表达水平,该效应在给药30min后达峰;而使用α7烟碱型乙酰胆碱受体特异性抑制剂--α银环蛇毒预处理细胞后,烟碱使其中磷酸化STAT5表达增高的效应消失。结论:烟碱可以增强CD4+CD25+调节性T细胞中磷酸化STAT5的表达,且该效应是依赖α7烟碱型乙酰胆碱受体途径的。
Objective:To identify the influence of α7 nicotinic acetylcholine receptor on STAT5 expression in mouse CD4+CD25+ regulatory T cells. Method:CD4+CD25+ regulatory T cells from the spleen of C57BL/6J mice were separated by means of MACS microbeads,and then we evaluated the STAT5 expression in these cells through western blot test at the present of receptor agonist and antagonist. Result:The findings show that nicotine,an agonist of α7 nicotinic acetylcholine receptor,could upregulate the p-STAT5 expression in regulatory T cells,and this effect peaks 30 minutes after drug administration. Pretreated cells with α7 nicotinic acetylcholine receptor's specific antagonist α-bungarotoxin,nicotine will no longer affect the p-STAT5 expression. Conclusion:Nicotine could enhance the expression of p-STAT5 in mouse CD4+CD25+ regulatory T cells,and this effect depends on the α7 nicotinic acetylcholine receptor pathway.
