摘要
目的研究妊娠期高血压疾病患者静脉血清及胎儿脐血中巨噬细胞移动抑制因子(MIF)和高迁移率蛋白-1(HMGB1)的变化及意义。方法采用酶联免疫吸附法(ELISA)进行MIF和HMGB1含量的测定,分别测定90例妊娠期高血压疾病患者(其中妊娠期高血压、轻度子痫前期、重度子痫前期各30例)及30例正常对照组产妇静脉血清及胎儿脐血中MIF和HMGB1的含量。结果 MIF和HMGB1在妊娠期高血压疾病组及对照组血清及胎儿脐血中均有表达。MIF在妊娠期高血压疾病各组血清中含量与对照组血清中的比较,差异有统计学意义(t=7.31,9.82,10.04,P值均<0.05),且重度子痫前期组明显高于轻度子痫前期和妊娠期高血压组(t=7.23,8.95,P值均<0.05);轻度子痫前期组明显高于妊娠期高血压组(t=7.68,P<0.05)。HMGB1在妊娠期高血压疾病各组血清中含量与对照组血清中的比较,差异有统计学意义(t=7.54,9.65,10.18,P值均<0.05),且重度子痫前期组明显高于轻度子痫前期和妊娠期高血压组(t=7.54,9.65,P值均<0.05);轻度子痫前期组明显高于妊娠期高血压组(t=7.35,P<0.05)。妊娠期高血压疾病患者MIF与HMGB1含量呈正相关(r=0.59,P<0.01)。MIF和HMGB1在妊娠期高血压疾病各组胎儿脐血中的含量,差异无统计学意义(P>0.05)。结论 MIF和HMGB1的高表达说明,妊娠期高血压疾病患者内皮细胞损伤,且MIF和HMGB1在损伤过程中起作用。
Objective To investigate the expression and significance of macrophage migration inhibitory factor(MIF) and high mobility group box-1(HMGB1) in intravenous serum and umbilical cord blood of patients with hypertensive disorders in pregnancy(pregnancy-induced hypertension syndrome,PIH).Methods The contents of MIF and HMGB1 in intravenous serum and umbilical cord blood from 90 patients with hypertensive disorders in pregnancy(which were divided into gestational hypertension,mild and severe preeclampsia groups each including 30 patients) were detected by enzyme-linked immunosorbent assay(ELISA).30 normal pregnant women were taken as control group.Results MIF and HMGB1 expressed in both patients and normal controls.Serum contents of MIF in patients and controls had significant difference(t=7.31,9.82,10.04,all P0.05).Moreover,there were differences among the contents of three groups(t=7.23,8.95,7.68,all P0.05).Serum contents of HMGB1 in patients and controls had significant difference(t=7.54,9.65,10.18,all P0.05).Moreover,there were differences among the contents of three groups(t=7.54,9.65,7.35,all P0.05).Contents of MIF and HMGB1 of patients with hypertensive disorders in pregnancy showed a positive correlation(r=0.59,P0.01).There was no significant difference in contents of MIF and HMGB1 in umbilical cord blood of patients with hypertensive disorders in pregnancy in three groups(P0.05).Conclusion The high expression of MIF and HMGB1 in patients with hypertensive disorders in pregnancy indicates the injury of vascular endothelium cells and MIF and HMGB1 play a role in the process of injury.
