摘要
为探索理想的外源性自由基致心律失常的模型,我们使用Langendorff灌流装置对大鼠离体心脏灌流9种不同浓度比的硫酸亚铁/抗坏血酸,同时用高效液谱法检测冠脉流出液中羟自由基(OH)的含量和观察心律失常的发生率。实验表明,硫酸亚铁(0.25mmol/L)/抗坏血酸(1.0mmol/L)生成的·OH含量较高,且使离体灌流大鼠的心律失常发生率高达100%,室性纤颤发生率为60%。可见,该系统是良好的外源性自由基致心律失常的模型。
In order to explore ideal model of arrhythmia induced by exogenous free radical generating system,we perfused the isolated rat heart with 9 different concentration of FeSo4/Vc using the Langendorff method .The amount of. OH in effluxion of the coronary artery were measured by HPLC, the incidence of arrythmias was recorded at the same time .The results of the experiment showed that the amountof .OH was higher,the incidence of arrhythmias was 100.0% and the incidence of VF was 60% Undercondition of exposure to FeSP4(0. 25mmol/L)Vc(1mmol/L) . It is obvious that this system is a best modelof arrhythmia induced by exogenous free radical generating sysytem.
