摘要
通过基因重组技术在完成粒/巨噬细胞集落刺激因子(GM-CS)高表达克隆的基础上对CM-CSF下游处理进行了较为系统的研究,得到了高纯度、高比活性的GM-CSF。本研究对其生物学活性进行了基础与临床试验,结果表明,rhCM-CSF对化疗后小白鼠及肿瘤患者白细胞降低有明显的升高作用,并对小白鼠及肿瘤患者因化疗引起的白细胞降低有明显的保护作用,为肿瘤患者临床应用GM-CSF提供了必要的方法与途径。
By using gene recombination techniques, we obtained a granulocyte -macrophage colony-shmulating factor (GM-CSF) with high expression and made systematic study on its purity and activity.According to the previous studies,the biological activities of GM-CSF in the mouse and patients were carried out in this paper. The results suggested that rhGM-CSF can increase the leukocyte remarkably in miceand patients after chemotherapy and can resist the decrease leukocyte caused by chemotherapy.
