摘要
目的通过免疫组化染色观察CD44和CD44v6在胚胎各脏器中的表达,了解这两种表面标志物在不同器官中的表达情况。方法用免疫组化PV-9000二步法(非生物素)检测CD44和CD44v6在6例胚胎各器官的表达情况。CD44用已知阳性的淋巴结作阳性对照;CD44v6用已知阳性的扁桃体作阳性对照。两者均用PBS代替一抗作阴性对照。结果 CD44主要表达于间叶组织中,如食管、胃及肠管的深肌层,心脏中的部分小血管、神经束以及各器官中的纤维组织,肝和脾内的造血细胞也有表达,但不表达于骨、肾、睾丸、心肌和肝细胞。另外,在少数上皮组织中也见有表达,如食管黏膜和气管黏膜上皮的基底层细胞及气管腺上皮。CD44v6的表达仅见于食管上皮基底层细胞中,其它各器官均未见表达。结论通过免疫组化法观察CD44和CD44v6在胚胎发育过程中的表达情况,为成体干细胞特性的研究提供了良好直观的依据和有用的线索。
Objective To study and compare surface markers in different organs of early embryos through detecting CD44 and CD44 variant 6(CD44v6) by immunohistochemical method in every organ of early embryos. Methods CD44 and CD44v6 in 6 cases of early embryos were detected by immunohistochemistry. Results CD44 was mainly expressed in mesenchymal tissues, such as deep muscle layer of esophagus and gut, part of vessels and nerve fiber in the heart , fibrous tissues in other organs, while it was negative in bone, kidney, testis, cardiac muscle and hepatic cells. In addition, CD44 was also expressed in a few of epithelium tissues, such as basilar membrane epithelial cells of esophagus, tracha and tracha gland. The weakly expression of CD44v6 was only observed in basal membrane epithelial cells of esophagus, other organs have not identified expression. Conclusion The special expression of CD44 and CD44v6 were observed in organs of early embryos by immunohistochemistry and the results will provide direct clues for the research of adults stem cells.
