超声法优化山楂叶中黄酮类成分的提取工艺 被引量:4

摘要 目的研究山楂叶中总黄酮的提取工艺,确立最佳提取条件.方法采用紫外检测法,建立山楂叶中黄酮类成分含量的测定方法,用正交设计法对影响超声提取的因素如甲醇浓度、甲醇用量、超声时间和提取次数进行考察,优化提取工艺.结果对山楂叶中黄酮类成分提取影响最显著的因素为甲醇用量.优化的提取条件为:加20倍量的80%的甲醇,连续提取两次,每次10 min.结论所建立的含量测定方法可用于山楂叶中黄酮类成分的含量测定,优化的提取工艺效率高,操作简单. Objective To optimize the extracting technology for flavonoid composions in leaves of Crataegus pinnatifida Bge.by orthogonal test.Method Flavonoid compositions in leaves of Crataegus pinnatifida Bge.was determined by UV detection.Effects on ultrasonic extraction of flavonoid compositions such as dosage of methanol,times of extraction,ultrasonic time and concentration of methanol were investigated by orthogonal design table,and the extraction technology was optimized.Result The most significant factor was the dosage of methanol.The optimized extraction conditions were: adding 20 times volume of 80% methanol with two successive extractions,and 10 minutes for each time.Conclusion The UV method established in the present study can be used to determine the content of flavonoid composition in leaves of Crataegus pinnatifida Bge.The optimized extraction method showed promise with higher extraction efficiency and simple processing.
