黔棘茎楤木对佐剂性关节炎大鼠治疗作用及机制探讨 被引量:11

Effects ofAralia echinocaulis in Guizhou on Adjuvant Arthritis in Rats and Related Mechanism

摘要 目的:观察黔棘茎楤木对佐剂性关节炎(AA)大鼠的作用,并探讨其作用机制。方法:设正常对照组、模型对照组、阳性药对照组(雷公藤多苷12 mg·kg-1)、黔棘茎楤木高、中、低剂量组(剂量分别为3.5,1.75,0.88 g·kg-1);除正常对照组外,其余各组均采用弗氏完全佐剂足跖sc复制AA模型;各组于造模当天ig给药,连续35 d;观察该药对AA大鼠原发性和继发性踝关节足肿胀及病理形态学影响;采用硝酸还原法检测各组血清中一氧化氮(NO)含量;放免法检测各组血清中白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量。结果:与模型组比较,黔棘茎楤木高剂量组可抑制AA大鼠继发性关节肿胀,抑制效果随剂量(中、低剂量组)降低而减轻。黔棘茎楤木高剂量组能显著降低AA大鼠血清中IL-1含量;高、中剂量组显著降低AA大鼠血清中TNF-αμg.L-1含量;高、中、低剂量组显著降低AA大鼠血清中NOμmol.L-1含量。病理学观察,黔产棘茎楤木高剂量组大鼠踝关节炎症细胞明显减少,滑膜组织细胞及成纤维细胞增生不明显,关节软骨破坏较少。结论:高剂量的黔棘茎楤木可以通过抑制炎症细胞因子,从而实现对AA大鼠的治疗作用。 Objective:To observe the effects of Aralia echinocaulis Hand-Mazz in Guizhou on adjuvant arthritis in rats and discuss the mechanism.Method: Rats were divided into normal control group,model group,positive drug group(tripterygium wilfordii 12 mg·kg-1),A.echinocaulis high dosage group,middle dosage group,low dosage group(the dosage 3.5,1.75,0.88 g·kg-1)randomly,all rats were made into ovalbumin adjuvant-induced arthritis model in Freund's adjuvant(FCA) except the normal group.The according treatments were given by oral gavage for each group starting from the modeling day,then continuously for 35 days.The effect on rats' primary and secondary adjuvant-induced arthritis were observed.Nitric oxide(NO) content in serum was detected by nitrate reductase method and the interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α) were measured with radioimmunoassay(RIA).Result:A.echinocaulis had obvious inhibition on rats' ankle adjuvant-induced arthritis,the inhibitory effect was decreased with the alleviation of the amount of dosage(middle dosage group,low dosage group).The A.echinocaulis high dosage group could decrease the IL-1 level in rat serum significantly.The high and middle dosage group could decrease the TNF-α level significantly.The high,middle and low dosage group could decrease the NO level significantly.From the pathological observation,the rat ankle joint inflammatory cells in A.echinocaulis high dosage group decreased obviously,the synovial membrane tissue cells and the fibroblast cells did not increase obviously,the articular cartilages got few failure.Conclusion: The high dosage A.echinocaulis is effective in the treatment of AA rats by inhibiting the inflammatory cytokines.
