小儿止咳颗粒对急性肺损伤小鼠高迁移率族蛋白B-1含量的影响 被引量:4

Effect of Xiaoer Zhike Granule on Serum High Mobility Group Box-1 Protein Level of Acute Lung Injury in Mice

摘要 目的:观察小儿止咳颗粒对急性肺损伤小鼠晚期炎症介质高迁移率族蛋白B-1(high mobility group box-1 protein,HMGB-1)含量的影响。方法:将ICR小鼠随机分成6组,分别测定空白组和脂多糖(LPS)尾iv致小鼠急性肺损伤后24,36,48,72,96 h血清中HMGB-1的含量,确定其含量最高的时间点。将ICR小鼠随机分为空白对照组、模型组、小儿止咳颗粒高、中、低剂量组,醋酸地塞米松组;除空白组外,各组尾iv LPS造成急性肺损伤模型,给药组分别ig给予小儿止咳颗粒5.4,2.7,1.35g·kg-1及醋酸地塞米松0.5 mg·kg-1,空白组ig给予等量生理盐水,在iv LPS 72 h测定各组小鼠血清中HMGB-1的含量。结果:iv LPS后72 h,小鼠血清中HMGB-1的含量最高,在该时间点,与模型组相比小儿止咳颗粒高、中、低剂量均能够抑制HMGB-1含量的增高(P<0.05)。结论:小儿止咳颗粒对晚期炎症介质HMGB-1含量的增高具有一定的抑制作用,这可能是其发挥治疗小儿上呼吸道感染后咳嗽作用的机制之一。 Objective:To observe the effect of Xiaoer Zhike Granule on serum high mobility group box-1 protein(HMGB-1)level in lung tissues of mice with endotoxin shock.Method:ICR mice were divided into six groups,except the blank group,all other groups were injected with LPS by the tails vein to establish the lung tissue injury model.The blank group was only received an injection of same volume of saline.The serum HMGB-1 level at 24,36,48,72 and 96 h was investigated respectively to explore the time of the highest content.ICR mice were divided into six groups as follows:blank,model,Xiaoer Zhike Granule high,middle,low group,dexamethasone acetate tablets.The effect of Xiaoer Zhike Granule on serum HMGB-1 content of sepsis mice was studied at the point of 72 h.Result:Serum HMGB-1 got to the highest level at 72 h after injection of LPS.Xiaoer Zhike Granule could inhibit the content's increase in HMGB-1(P0.05).Conclusion:Xiaoer Zhike Granule can decrease serum HMGB-1' content,which maybe related to its function.
