摘要
对橡胶籽中β-葡萄糖苷酶活力测定的条件进行了探讨。结果表明最适测定条件为:0.1mL适度稀释粗酶液,使反应最终吸光值在0.1~1.0范围内,0.3mLpH6.0磷酸二氢钾-磷酸氢二钠缓冲液,0.1mL10mmol/LpNPG体系,在42℃下反应30min,于400nm测定对硝基苯酚含量。按照此方法测得橡胶籽中β-葡萄糖苷酶活力为:105.632U/g,该方法相对标准偏差为:2.017%,精密度较好。
The optimum assay condition of β-glucosidase activity in Hevea brasiliensis seeds was investigated. The results suggest that enzyme activity is assayed by 0.1 mL crude β-glucosidase, 0.3 mL Na2HPO4-KH2PO4 buffer(pH 6.0) and 0.1 mL 10 mmol/L pNPG at 42 ℃ for 30 min, then the reaction product p-nitrophenol is measured by spectrophotometer at 400 nm. The β-glucosidase activity in Hevea brasiliensis seeds is determined as 105.632 U/g, with RSD 2.017%.
