金匮肾气丸对聋豚鼠耳蜗GAP-43、NGF的影响

Effect of Jinkui Shenqi Pills on Growth Associated Protein-43 and Nerve Growth Factor in the Cochlea of Deafness Guinea Pigs

摘要 目的:探讨金匮肾气丸对庆大霉素造成药物性耳聋豚鼠耳蜗螺旋神经节神经修复相关蛋白表达的影响。方法:30只豚鼠随机分成3组各10只,即正常组、模型组、治疗组。将模型组豚鼠按100mg/kg/d注射庆大霉素,治疗组在注射庆大霉素后灌服金匮肾气丸水煎剂,实验结束后取耳蜗标本,免疫组化观察其生长相关蛋白(GAP-43)及神经生长因子(NGF)的表达。结果:GAP-43、NGF在实验动物耳蜗螺旋神经节细胞中都有一定的表达;正常组表达较弱较少,其它各组表达均较正常组表达增加,其中治疗组表达高于模型组(P<0.01)。结论:金匮肾气丸可提高庆大霉素致聋豚鼠耳蜗GAP-43、NGF表达,对螺旋神经节细胞进行修复。 Objective:To explore the effect of Jinkui Shenqi Pills on the expression of nerve repair protein in cochlear ganglion of drug-induced deafness guinea pigs.Methods:Thirty guinea pigs were equally divided into three groups:the normal control group,the model group and the treatment group.The model group and the treatment group were given intramuscular injection with gentamicin 100mg/kg/d.The treatment group were given Jinkui Shenqi Pills after injection of gentamicin.After experiment,Nerve growth factor(NGF) and growth associated protein 43(GAP-43) expressions in the cochlea were detected by immunohistochemistry.Results:GAP-43 and NGF had certain expression in the spiral ganglion cells of cochlea of the tested animals.But the expression was weak in the normal control group.The treatment group had obvious expression of GAP-43 and NGF(P 〈 0.01 compared with the model group).Conclusions:Jinkui Shenqi Pills can restore the injured spiral ganglion cells by increasing the expression of GAP-43 and NGF in the cochlea of gentamicin-induced deafness guinea pigs.
