摘要
目的:观察痛必定注射液对大鼠脊髓背根神经节(DRG)细胞N-甲基-D-天门冬氨酸2B亚型受体(NR2B)含量的影响,以探讨其外周镇痛的机理。方法:将40只Wister大鼠分为A(正常组)、B(假手术组)、C(模型组)组、D(模型+痛必定组)4组各10只,采用坐骨神经结扎慢性损伤动物模型,通过免疫组化技术观察痛必定注射液对于DRG细胞内NR2B含量的影响。结果:A、B组NR2B含量比较,差异无显著性意义(P>0.05);C组NR2B含量较A、B组明显升高(P<0.05);D组NR2B含量较C组明显降低(P<0.05),与A、B组比较,差异无显著性意义(P>0.05)。结论:痛必定注射液的外周镇痛作用可能是通过抑制NR2B的合成或释放而实现的,由此可推断NR2B与"不通则痛"的病机相关。
Objective:To observe the effect of Tongbiding Injection on N-meth-D-aspartic acid receptor subtypes 2B(NR2B) in the dorsal root ganglion(DRG) of rats with chronic injury induced by constriction of sciatic nerve.Methods:Forty Wister rats were randomly divided into the control group(group A),the sham-operation group(group B),the model group(group C),and Tongbiding Injection group(group D),ten rats in each group.Chronic injury in rats was induced by constriction of sciatic nerve.The changes of NR2B in rats DRG were detected by immunohistochemistry.Results:There was no significant difference of NR2B level between groups A and B(P 〉 0.05).NR2B level of group C was higher than that of groups A and B(P 〈 0.05),while NR2B level of group D was lower than that of group C(P 〈 0.05).There was no significant difference among groups A,B and D(P 〈 0.05).Conclusion:The peripheral analgesic mechanism of Tongbiding Injection is probably related with the inhibition of synthesis or release of NR2B in rats DRG,indicating that NR2B has certain relation with the pathogenesis of obstruction resulting in pain.
