抗癌生物活性肽对实验性裸鼠肿瘤组织中p53和Bcl-2表达的影响

摘要 目的探讨抗癌生物活性肽(ACBP)作用于荷胃癌裸鼠后对肿瘤中p53和Bcl-2蛋白表达的影响,探讨ACBP可能的作用机制。方法成功建立荷胃癌裸鼠模型后将鼠随机分为2组,各组10只。对照组给予生理盐水;实验组腹腔注射活性肽0.4 mL(50 mg/L)。连续处理35 d,停药4 d后处死裸鼠,取肿瘤,应用免疫组化链霉亲和素-生物素-过氧化物酶复合物法检测突变型p53蛋白及Bcl-2蛋白的表达情况。结果 p53和Bcl-2蛋白在对照组肿瘤中阳性表达较强,在活性肽组中的表达均比对照组的表达弱,两组比较差异有统计学意义(P<0.01)。结论 ACBP对荷瘤裸鼠肿瘤生长有明显的抑制作用,可能是通过影响p53和Bcl-2的表达实现的。 Objective To investigate the expression difference of apoptotic related gene p53 and Bcl-2 of anti cancer bioactive peptide(ACBP) treated tumor-loaded nude mice and the potential anti cancer mechanism of ACBP.Methods Nude mice were divided into 2 groups randomly: 10 as control group and 10 as experimental group.2 groups of nude mice were administered with 0.4 mL physiological saline,0.4 mL ACBP(50 mg/L)continuously for 35 days,respectively.Nude mice were executed after the administration finished 4 days later,then,tumors were taken out.Then,p53 and Bcl-2 expression were detected using immunohistochemistry strept actividin-biotin complex(SABC) method between groups.Results The expression of p53 and Bcl-2 was relatively stronger in control group than that in experimental groups and which showed significant difference(P〈0.01).Conclusion Anti cancer bioactive peptide can obviously inhibit the nude mice sarcoma.The mechanism may be correlated with the decreased expression of p53 and Bcl-2 protein.
