RP-HPLC法测定丹参不同部位中丹参素和原儿茶醛的含量 被引量:6

摘要 目的采用RP-HPLC法测定丹参不同部位中丹参素和原儿茶醛的含量。方法色谱柱为Kromasil C18柱(150 mm×4.6mm,5μm),流动相为甲醇-乙腈-1%冰醋酸(7∶3∶90),流速1.0 ml.min-1,检测波长280 nm,柱温为室温。结果丹参素和原儿茶醛的线性范围分别为2.333~30.000 mg.L-1(r=0.999 7)和0.397~5.100 mg.L-1(r=0.999 2);平均回收率分别为100.6%(RSD=1.43%)、100.8%(RSD=1.37%)。结论该方法简便、准确、重复性好。丹参的茎和叶中含有一定量的丹参素和原儿茶醛。 Aim To develop a method for determination of danshensu and protocatechuic aldehyde from different parts of Salvia miltiorrhiza Bunge by RP-HPLC.Methods A separation was performed on a Kromasil C18 column(150 mm×4.6 mm,5μm) at room temperature.The mobile phase was methanol-acetonitrile-1% acetic acid glacial(7∶ 3∶ 90).The flow rate was 1.0 ml·min-1 and the detective wavelength was at 280 nm.Results The results showed that the quantitative linerange of danshensu and protocatechuic aldehyde were from 2.333 to 30.000 mg·L-1(r=0.999 7)and from 0.397 to 5.100 mg·L-1(r=0.999 2),respectively.The average recovery rate of danshensu and protocatechuic aldehyde was 100.6%(RSD=1.43%) and 100.8%(RSD=1.37%),respectively.Conclusion This method is simple,accurate and reproducible.There are danshensu and protocatechuic aldehyde in stem and leaves of Salvia miltiorrhiza Bunge.
