摘要
目的研究直肠癌中Slit2/Robo1蛋白的表达,探讨两者与直肠癌微血管密度(MVD)及生物学行为的关系。方法免疫组化SP法检测65例直肠癌组织及相应43例对照组织中Slit2/Robo1蛋白的表达水平及MVD计数,研究在直肠癌中Slit2/Ro-bo1蛋白的表达强度与MVD关系,观察不同临床病理学行为下Slit2/Robo1蛋白的表达变化。结果(1)直肠癌组织中Slit2/Robo1蛋白阳性表达率分别为64.6%和78.5%,对照组织中的阳性表达率分别为4.7%和7.0%,两种组织中表达差异有统计学意义(P<0.01),直肠癌组织中MVD显著增高(t=32.09,P<0.01);(2)Slit2与Robo1阳性表达呈正相关(r=0.551,P<0.01),两者表达阳性的癌组织MVD计数明显大于表达阴性的癌组织(t=5.674,P<0.01,t=6.103,P<0.01);(3)Slit2/Robo1蛋白表达水平与肿瘤大小、浸润深度、Dukes分期及淋巴结转移相关,统计学检验有显著性差异(P<0.05)。结论(1)Slit2/Robo1蛋白在直肠癌组织中表达较对照组明显增高,并与MVD密切相关,推测Slit2/Robo1蛋白在直肠癌血管生成中起着重要作用;(2)直肠癌组织中Slit2/Robo1蛋白及MVD与直肠癌的生长、侵袭及转移密切相关,临床通过检测Slit2/Robo1蛋白表达强度及MVD计数可作为直肠癌进展程度及预后情况的重要指标。
Aim To investigate the expression of Slit2/Robo1 proteins in human rectal carcinomas and their correlation with microvascular density and biological behaviors.Method The expression of Slit2/Robo1 proteins and CD105-labeled microvascular density were measured by immunohistochemical staining(SP) in 65 cases of rectal carcinomas and 45 cases of control tissues group.Result The positive rates of Slit2/Robo1 were 64.6% and 78.5% in rectal carcinomas,4.7% and 7.0% in control tissues group,and the expression of Slit2/Robo1 and MVD in cancerous tissues were higher than those in control tissues group(P0.01).There was a significant positive correlation between Slit2/Robo1 expression(r=0.551,P0.01).The MVD in the tumor tissues with positive expression of Slit2/Robo1 was much higher than that in the negative one(P0.01).The expression rates of Slit2/Robo1 were significantly associated with tumor size,invasion depth,Dukes stage,lymph node and distant metastasis.Conclusion Slit2/Robo1 signaling were distinctively over-expressed and might play an important role in the angiogenesis of rectal cancer.The Slit2/Robo1 proteins and angiogenesis play an important role in cancer invasion and metastasis.The expression intensity of Slit2/Robo1 and the enumerations of MVD can be used an important index for the capacity of growth,invasion and metastasis in the rectal carcinomas
