微量刃天青显色法快速测定结核分枝杆菌MIC及菌型鉴定 被引量:5

摘要 目的用刃天青显色法快速测定结核分枝杆菌(MTB)MIC及菌型初步鉴定,并确定抗结核药物折点浓度。方法用30株国家结核参比实验室熟练度测试质控菌株和64株临床分离菌株为试验菌株,在96孔培养板中,用Middlebrook 7H9液体培养基将抗结核药物进行连续双倍稀释后,加入一定浓度菌液,用刃天青显色法测定MIC。根据罗氏培养基药敏比例法结果,用ROC曲线分析建立显色法药物折点浓度。同时选择24株非结核分枝杆菌(NTM),用对硝基苯甲酸(PNB)进行初步的菌型鉴定。结果刃天青显色药敏试验5种抗结核药物链霉素、异烟肼、利福平、乙胺丁醇和氧氟沙星的折点浓度分别为1、0.5、0.5、2、2μg/mL,其敏感性分别为100%、97%、97%、88%和100%,特异性分别为100%、100%、100%、91%和100%。在菌型鉴定中,94株MTB在PNB中均未生长,24株NTM均生长。结论刃天青显色法测定MIC具有操作简便、快速、成本低廉和准确的优势,且能进行初步的菌型鉴定,适于结核病专业实验室应用。
