摘要
目的探讨凋亡抑制基因livin在卵巢癌细胞中的表达及其生物学意义。方法收集32例卵巢癌患者癌组织及相应的癌旁组织、细胞培养卵巢癌细胞系A2780,用实时荧光定量PCR和westernblot分别检测组织中livin基因的mRNA和蛋白质表达;MTT法检测A2780细胞的生长率;流式细胞术检测A2780细胞的凋亡率。结果 livinmRNA和蛋白质表达在卵巢癌组织中明显高于癌旁组织(t=6.98,P<0.01;t=5.12,P<0.01);与对照组相比,转染livinsiRNA组卵巢癌细胞的生长率显著减少(χ2=77.21,P<0.01),细胞凋亡率呈明显增高(χ2=15.68,P<0.01)。结论 livin在卵巢癌的发生、发展中起重要的作用,可能成为卵巢癌治疗的新分子靶点。
Objective To investigate the expression and the biological significance of the apoptosis suppressor genes livin in ovarian cancer cells.Methods The primary tumor tissues,matching adjacent non-cancerous tissues from ovarian cancer and ovarian cancer cell line A2780 were collected.The expression of livin was examined by real-time RT-PCR.Livin protein levels were detected by western blot.The growth rates of A2780 cell line were detected by MTT method and the apoptosis rates were detected by flow cytometric technology.Results The expression of livin in ovarian tissues was significantly higher than that in adjacent non-cancerous tissues (t=6.98,P 0.01;t=5.12,P 0.01),Compared with the control group,the growth rate of ovarian cancer cells which carried with livin siRNA was significantly reduced (χ2=77.21,P 0.01) and its apoptosis rate was significantly increased (χ2=15.68,P 0.01).Conclusion Livin may play an important role during ovarian cancer occurrence and development,and become a new molecular target for the treatment of ovarian cancer.
