摘要
目的探讨地黄管食通口服液对食管癌放疗大鼠模型的影响及作用机制。方法 Wistar大鼠20只为空白组,110只皮下注射甲基戊基亚硝铵5mg/kg建立食管癌模型。除食管癌组20只外,其余分为5组,均应用60Co食管局部放疗。单纯放疗组(单放组)18只,其余分成六味组(18只,浓度0.45g/ml六味地黄丸混悬液放疗后灌胃10ml/kg),地黄管食通高、中、低剂量组(各18只,浓度分别为1.5g/ml、1.0g/ml、0.5g/ml地黄管食通口服液放疗后灌胃10ml/kg)。空白组、食管癌组、单放组每天灌服蒸馏水10ml/kg,各组均灌胃5周。镜下观察各组大鼠食管端粒酶逆转录酶(hTERT)形态学及阳性率比较。结果镜下地黄管食通高、中、低剂量组及六味组胞核及细胞浆呈棕黄色着染,着色明显比食管癌组浅。与食管癌组比较,各药物组hTERT阳性率差异均有统计学意义(P<0.01);高剂量组与单放组比较差异有统计学意义(P<0.05);各药物组之间比较差异无统计学意义(P>0.05)。结论地黄管食通口服液可能通过下调hTERT活性表达从而防治食管癌放疗后复发。
Objective To discuss the effect and functional mechanism of Dihuang Guanshitong Oral Liquid(Oral liquid of Radix Rehmanniae for esophageal cancer) on esophageal cancer model rats after radiotherapy.Methods Twenty Wistar rats were put into a blank group.Another 110 rats were given subcutaneous injection of methyl amyl ammonium nitrate 5mg/kg to establish esophageal cancer models.Except the 20 rats in the esophageal cancer group,the rest of rats were divided into five groups and treated by 60Co radiotherapy for the local area.There were 18 rats in the simple radiotherapy group.The others were divided into Liuwei group [18 rats,Liuwei Dihuang Wan(Bolus of Six Medicines Including Rehmanniae) suspension in the concentration of 0.45g/ml for intragastric feeding after radiotherapy],Dihuang Guanshitong high-dosage,medium-dosage,and low-dosage groups(18 rats in each group,Dihuang Guanshitong Oral Liquid in the concentration of 1.5g/ml,1.0g/ml,and 0.5g/ml respectively for intragastric feeding 10ml/kg after radiotherapy).The blank group,esophageal cancer group,and simple radiotherapy group were treated by intragastric feeding with distilled water 10ml/kg for 5 weeks.The morphological changes and the positive rate in the esophageal human telomerase reverse transcriptase(hTERT) of all groups were observed with microscope.Results The nucleus and cytoplasm of the Dihuang Guanshitong high-dosage,medium-dosage,and low-dosage groups and Liuwei group were in the brown-yellow colour in staining,significantly light-coloured than that of the esophageal cancer group.Compared with the esophageal cancer group,the difference in the hTERT positive rate of each medicated group was all statistically significant(P0.01).The difference between the Dihuang Guanshitong high-dosage group and simple radiotherapy group was statistically significant(P0.05).There was no statistical difference in the comparison among all medicated groups(P0.05).Conclusion Possibly,Dihuang Guanshitong Oral Liquid prevents the recurrence of esophageal cancer after radiotherapy through down-regulating the activity of hTERT expression.
