摘要
目的探讨天然冰片及其结构修饰物对戊四唑所致惊厥模型小鼠氨基酸类神经递质含量的影响。方法采用戊四唑制备小鼠惊厥模型,观察给药后动物惊厥的发生及其潜伏时间,再通过高效液相色谱-荧光检测法测定脑内氨基酸类神经递质的含量。结果与模型组比较,天然冰片高剂量组小鼠抗惊厥数量显著增加,惊厥潜伏期显著延长;(+)-顺丁烯二酸单冰片酯中剂量组惊厥潜伏期显著延长。与模型组比较,天然冰片低剂量组脑内天冬氨酸(ASP)显著增加、谷氨酸(Glu)含量显著减少;天然冰片高剂量组γ-氨基丁酸(GABA)含量显著增加,(+)-顺丁烯二酸单冰片酯中剂量组Glu含量显著减少。结论天然冰片及其结构修饰物可能通过改变脑内氨基酸类神经递质的含量来改善模型小鼠的惊厥行为。
Objective To study the effect of natural borneol and its structural modifiers on cerebral contents of amino acid neurotransmitters in mice with pentylenetetmzol-induced convulsion.Methods Pentylenetetmzol-induced convulsion mice were used to observe the effect of borneol and its structural modifiers on the incidence and the latent period of convulsion.After treatment,mice brain was taken out to determine the amino acid neurotransmitters contents by HPLC-fluorescence detection method.Results Compared with the model group,high-dose natural borneol group increased the number of anticonvulsant and prolonged the latent period of convulsion significantly(P 0.05),and medium-dose(+)-bornyl monomaleate group also prolonged the latent period of convulsion significantly(P 0.05).Compared with the model group,the content of aspartate(ASP) was increased significantly and the content of glutamate was decreased significantly in mice brain of low-dose natural borneol group(P 0.05),and the content of γ-aminobutyric acid was increased significantly in the high-dose natural borneol group(P 0.05);the content of glutamate was decreased significantly in the medium-dose group of(+)-bornyl monomaleate(P 0.01~0.05).Conclusion Natural borneol and its structural modifiers improve the convulsion behavior of the model mice probably through regulating the content of the amino acid neurotransmitters in mice brain.
