摘要
目的研究葛根素注射液对Beagle犬红细胞的溶血作用。方法取24只Beagle犬的红细胞,分别与不同浓度的葛根素注射液(1~10 mg/mL)、丙二醇溶液(2%~20%)共同孵育,观察24 h内红细胞的状态和溶血发生率,用χ2检验统计分析溶血实验结果。结果增大葛根素注射液浓度、延长与供试品(红细胞)反应时间(葛根素10 mg/mL,丙二醇浓度20%,反应时间24 h),24只Beagle犬红细胞的溶血发生率高达66.7%,而等浓度丙二醇对照溶液组的溶血发生率为16.7%,两者差异有显著性(P<0.01)。结论与常规溶血试验方法相比,本研究中改进的溶血反应评价方法能够更准确检测出葛根素注射液所致的偶发性溶血反应,所致溶血反应主要由高浓度的葛根素所引起。
Objective To investigate the hemolysis induced by Puerarin Injection(PI) in red blood cell(RBC) of Beagle dogs.Methods Red blood cells were collected from 24 Beagle dogs separately.And then RBC were incubated with different concentrations of puerarin solution(1~10 mg/mL) and 1,3-propanediol solution(2~20 %) for 24 hours.The changes of RBS and hymolysis incidence were observed,and the results were analyzed with chi-squared test.Results PI solution at high concentration(puerarin of 10 mg/mL,propanediol of 20 %) caused obvious hemolysis with an incidence of 66.7 %.In the 20 % propanediol-treated group,the hemolytic incidence was 16.7 % which was significantly lower than that in the PI-treated group(P 0.01).Conclusion To detect the accidental PI-induced hemolysis in vivo,it is recommended to increase PI solution concentration,the amount of blood sample and the incubation time.High concentration of puerarin is the main cause in PI-induced hemolysis.
