温下肠腑方对大鼠大肠癌细胞周期素D1、增殖细胞核抗原表达的影响

Effects of Wenxiachangfu Formula on the Expression of Cyclin D1,PCNA in Rats with Colorectal Cancer

目的探讨温下肠腑方对二甲基肼诱导的大鼠大肠癌细胞周期素D1(CyclinD1)、增殖细胞核抗原(PCNA)表达的影响。方法予以大鼠1,2-二甲基肼腹腔注射,建立大鼠大肠癌模型,温下肠腑方灌胃给药干预,在第15、20、23周时间点,分别用免疫荧光组织化学法结合激光共聚焦显微镜扫描和免疫组织化学法观察中药对大鼠肠组织CyclinD1和PCNA表达的影响。结果在第15、20、23周时间点,与对照组比较,模型组大鼠肠组织CyclinD1蛋白和PCNA蛋白表达均显著增高,差异有统计学意义(P<0.05或P<0.01);与模型组比较,温下方组Cyclin D1蛋白和PCNA蛋白表达显著降低,差异有统计学意义(P<0.05)。结论温下肠腑方能够显著降低肠癌动物模型肠组织Cy-clinD1、PCNA表达,提示温下肠腑方可能通过抑制CyclinD1、PCNA的表达而抑制肠癌细胞的增殖。

Objective To discuss the effects of wenxiachangfu formula on the expressions of cyclin D1 and proliferating cell nuclear antigen（PCNA）in the rats with colorectal cancer induced with dimethyl hydrazine.Methods The intraperitoneal injection with 1,2 dimethyl hydrazine（DMH）was applied to establish the model of colorectal cancer in rat.The medicinal intervention of wenxiachangfu formula was administrated through gastric infusion.On the time points of the 15th,20th and 23rd weeks,the immunofluorescene histochemical method combined with confocal laser scanning microscopy and immunohistochemical method were used to observe the effects of Chinese herbal medicine on the expressions of cyclin D1 and PCNA in colorectal tissue of rats.Results On the time points of the 15th,20th and 23rd weeks,compared with control group,the expressions of cyclin D1 protein and PCNA protein in the colorectal tissue of rat increased significantly in model group,presenting statistical significant difference（P0.05 or P0.01）.Compared with model group,the expressions of cyclinD1 and PCNA in wenxiachangfu formula decreased signficiantly,indicating significant difference（P0.05）.Conclusion Wenxiachangfu formula significantly reduces the expressions of cyclin D1 and PCNA in the colorectal tissue of the animal model with colorectal cancer.It is proposed that wenxiachangfu formula inhibits colorectal cancer cellular proliferation by down-regulating the expressions of cyclin D1 and PCNA.
