益肾降浊冲剂对慢性肾功能衰竭大鼠肾小管上皮细胞凋亡的影响 被引量:4

Effect of Soluble Granules Yishenjiangzhuo on Renal Tubular Epithelial Cells Apoptosis in Chronic Renal Failure Rats

摘要 目的观察益肾降浊冲剂对慢性肾功能衰竭(CRF)大鼠肾小管上皮细胞凋亡的影响,探讨其延缓CRF进展的作用机制。方法采用5/6肾切除建立CRF大鼠模型,随机分为假手术组、模型组、氯沙坦组、益肾降浊冲剂组,建模2周后,氯沙坦组、益肾降浊冲剂组分别予氯沙坦和益肾降浊冲剂灌胃6周,PAS染色观察肾小管间质的损伤程度,Tunel染色检测肾小管上皮细胞凋亡。结果模型组残肾肾小管出现萎缩、扩张和间质纤维化,Tunel阳性细胞数明显高于假手术组(P<0.01);与模型组比较,氯沙坦组、益肾降浊冲剂组肾小管间质损害明显减轻,Tunel阳性细胞数显著减少(P<0.01);益肾降浊冲剂组Tunel阳性细胞数明显低于氯沙坦组(P<0.01)。结论益肾降浊冲剂能抑制CRF残肾小管上皮细胞的凋亡,改善肾小管间质的损伤,疗效优于氯沙坦。 Objective To observe the effect of Soluble Granules Yishenjiangzhuo on renal tubular epithelial cells apoptosis in chronic renal failure (CRF) rats, and explore its mechanism. Methods The models of CRF rats were established by 5/6 subtotal nephrectomy, and the rats were randomly divided into sham operation group, model group, Losartan group and Soluble Granules Yishenjiangzhuo group. Two weeks after nephrectomy, Losartan group and Soluble Granules Yishenjiangzhuo group received intragastric administration of Losartan, Soluble Granules Yishenjiangzhuo respectively for 6 weeks. The renal tubular interstitial darnage was observed with PAS staining, and the apeptosis of renal tubular epithelial cells was detected with TUNEL staining. Results Renal tubular atrophy, dilation and interstitial fibrosis were seen in model group, and the number of positive cells of TUNEL was significantly higher than that of model group (P〈0.01). Compared with model group, the renal tubular interstitial damage was alleviated and the number of positive cells of TUNEL was reduced in Losartan group and Soluble Granules Yishenjiangzhuo group (P〈0.01). The number of positive cells of TUNEL in l_osartan group was lower than that of Soluble Granules Yishenjiangzhuo group (P〈 0.01). Conclusion Soluble Granules Yishenjiangzhuo can inhibit the apoptosis of renal tubular epithelial cells, alleviate the renal tubular interstitial damage, and its effects is better than that of Losartan.
