摘要
目的观察复方丹参滴丸对血管性痴呆(VD)大鼠的学习记忆能力影响并探讨其作用的机制。方法采用双侧颈总动脉结扎法制作血管性痴呆大鼠模型,用复方丹参滴丸治疗,Morris水迷宫检测大鼠的学习记忆能力,HE染色观察海马的病理变化,免疫组织化学检测海马CA3区Bax和Caspase-3的表达。结果复方丹参滴丸能明显改善VD大鼠的学习记忆能力,减轻VD大鼠海马的病理损害,减少VD大鼠海马CA3区Bax和Caspase-3的表达。结论复方丹参滴丸通过抑制海马CA3区Bax和Caspase-3表达,保护海马细胞,改善VD大鼠的学习记忆能力。
Objective To observe the effect of compound Danshen dripping pill (CDDP) on ability of learning and memory in rats of vascular dementia (VD) and explore its mechanism. Methods The models of VD rats were established by ligation of the bilateral common carotid artery and treated with CDDP. The ability of learning and memory of rats was detected by Morris water maze, pathological change of hippoeampus was observed by HE staining, and expressions of Bax and Caspase-3 in hippocampal CA3 region were detected by immunohistochemistry. Results The ability of learning and memory of rats in treatment group was higher than that of model group, and the hippocampal pathological damage in treatment group was slight, while the lesions in model group was more severe. The expressions of Bax and Caspase-3 in hippocampal CA3 region in treatment group were lower than that of model group. Conclusion CDDP can improve the ability of learning and memory of VD rats and protect hippocampal pyramidal cells by inhibiting the expressions of Bax and Caspase-3 in hippocampal CA3 region.
